HI Chris
You can test voltages anytime you want. Just be very careful and make sure any wires that aren't connected can't touch anything.
I like to check all the AC voltages as soon as the primary side of the power transformer is connected. I also put in the rectifier tube and check the DC voltages as soon as the board is connected.
The voltages will read high with no load, but at least you know the power supply is working.
