The TMB channel on the sIII V6 seems to have suddenly lost all its guts. There still is some gain, but it's really marginal; like wide-open gain and volume is what a quarter or less used to be. The normal channel works fine. The boost circuit works, and does boost the TMB, but only as much as you would expect for the gain on tap.
Any ideas where I should start looking?

Geez, what a novice- bad tube!.
I but the amp on the bench and checked voltages and found the bad spot at pin 8 on V2. Change the tube, and all is good.
Is there an easier way to do this or what? Do you just start recognizing the typical tube problems?

Well, glad you found the problem. :D

Swapping tubes is always a plan. You will get used to the sound when tubes are going bad, microphonic etc.

In this case, if you switched to the other set of tubes, you'd know it was them since the other set would [probably] work fine.
